Abstract

The upcoming revision of ICH Q6(R1), the International Council for Harmonisation (ICH) guideline on specifications, represents an important step towards aligning quality expectations for both chemical and biological medicines. Evolving from the existing Q6A and Q6B guidelines, the new document aims to bring these principles together into a single, practical framework that reflects current scientific knowledge and regulatory experience.

ICH Q6(R1) is expected to establish a harmonised approach to defining, justifying and implementing specifications across diverse product classes, including complex biologics and advanced therapies. The revision seeks to ensure that specification-setting remains aligned with advances in analytical capabilities and quality-by-design principles, while maintaining a clear focus on patient safety and product performance.

This harmonisation framework supports the wider ICH goal of promoting science- and risk-based regulatory strategies. By updating and integrating the previous guidelines, ICH Q6(R1) is expected to simplify global expectations, enhance flexibility and strengthen consistency in pharmaceutical quality standards.

This article provides an overview of the key concepts and anticipated impacts of ICH Q6(R1), highlighting how the integrated approach may reshape expectations for both traditional and innovative pharmaceutical products.
